Tissue specific RNA-Seq

Spores of C. richardii (RNWT1 strain) were spread and germinated on 1/2 Murashige & Skoog (MS) medium without sucrose at 26 °C under a 16-h light/8-h dark photoperiod, and sterilized water was added once a week. It consists of five tissues: leaf tip, leaf blade, shoot tip, root tip, and mature root region.

2,4-D induce RNA-Seq

For chemical treatments, 2 μM 2,4-D (Sigma, Cat. 573760, dissolved in DMSO), were added to 1/2 MS medium. Plants were grown vertically on 1/2 MS medium for chemical treatments. Mock controls were performed using the corresponding solvent.

KT induce RNA-Seq

For chemical treatments, 10 μM KT were added to 1/2 MS medium. Plants were grown vertically on 1/2 MS medium for chemical treatments. Mock controls were performed using the corresponding solvent.
