Arabidopsis seeds of wild-type Col-0 were germinated on 1/2 MS medium with 1% sucrose at 22 ˚C under a 16-h light/8-h dark photoperiod. The first pair of rosette leaves were detached from 12-day-old seedlings and cultured on sucrose-free B5 medium. The wounded regions of leaf explants at 4 d after detachment were collected for single-cell extraction and RNA-seq analysis. Cell-subclusters were analyzed using cells from cell-clusters 1, 12, 16, 17.
